Correct Grammar

Could you please tell me if the following sentence is grammatically correct?

 

By being able not only to understand one’s mistakes, but also to use them to improve one's own knowledge and ability is one of the defining characteristics of a great leader.

I'm not sure if there is supposed to be a comma between 'ability' and 'is'.

By in the beginning is not required.No comma is required after ability.
